Agnusides are iridoid glycosides found primarily in Vitex agnus-castus (chasteberry), a medicinal plant traditionally used for women's reproductive health. The use of agnusides or Vitex agnus-castus to support the dopaminergic system is rooted in historical and traditional medicine, particularly for conditions such as premenstrual syndrome (PMS), menstrual irregularities, and mastalgia. The proposed mechanism—supported by some preclinical and limited clinical data—is that constituents of Vitex, including agnusides, may have dopaminergic activity, acting as dopamine receptor agonists in the pituitary gland. This dopaminergic effect is thought to suppress prolactin secretion, which can be beneficial for certain menstrual disorders.
However, direct scientific evidence specifically linking agnusides to comprehensive dopaminergic system support across the body is limited. Most research focuses on Vitex extracts as a whole rather than isolated agnusides, and the primary outcome measures are related to prolactin levels and reproductive health parameters, not broad dopamine system modulation.
