Vitamin B12 (cyanocobalamin) is scientifically studied for its role in mental health, including depression. Deficiency in vitamin B12 is associated with neuropsychiatric symptoms such as mood disturbances, cognitive decline, and, in some cases, depression. Several observational studies have found that individuals with low levels of B12 are more likely to experience depressive symptoms. The proposed mechanisms involve B12’s role in methylation, neurotransmitter synthesis (such as serotonin and dopamine), and homocysteine metabolism, all of which are important for brain function and mood regulation.
However, the evidence for vitamin B12 supplementation as a treatment for depression is mixed. Supplementation appears beneficial primarily in people who are deficient in B12; in such cases, correcting the deficiency can alleviate depressive symptoms. Randomized controlled trials (RCTs) have not consistently shown benefit in non-deficient populations, and meta-analyses suggest only a modest effect, mainly as adjunctive therapy. Major treatment guidelines do not recommend B12 supplements as a primary therapy for depression, but they do recommend screening for and correcting deficiencies in at-risk populations (e.g., older adults, vegetarians, those with malabsorption).
In summary, scientific evidence supports checking and correcting B12 deficiency in depressed patients, but B12 is not a stand-alone treatment for depression in otherwise healthy individuals with normal levels.
