Mannanoligosaccharides (MOS) are prebiotic compounds derived from the cell walls of yeast, primarily Saccharomyces cerevisiae. Their use in supporting or mitigating the side effects of antibiotics is supported by some scientific evidence, though most of the data comes from animal studies rather than human clinical trials. Antibiotics can disrupt the balance of gut microbiota, leading to gastrointestinal side effects such as diarrhea. MOS are believed to help restore gut microbial balance by selectively promoting the growth of beneficial bacteria, such as Bifidobacteria and Lactobacilli, and by inhibiting the adhesion of pathogenic bacteria to the intestinal lining, which can decrease the risk of antibiotic-associated diarrhea. Several studies in poultry, swine, and other animals have demonstrated that MOS supplementation can help maintain gut health during and after antibiotic treatment, reducing the incidence of digestive disturbances. In human health, the evidence is more limited but suggests a similar mechanism may be present. Some small-scale studies and reviews indicate that MOS could have a positive effect in maintaining gut barrier function and microbial diversity during antibiotic therapy, but large-scale, high-quality clinical trials are lacking. Therefore, while the theoretical basis and animal data are promising, the direct evidence for MOS supporting or treating antibiotic side effects in humans is currently limited, justifying a moderate evidence rating.
