Vitamin B5 (Pantothenic Acid) is essential for the synthesis of coenzyme A, which is necessary for fatty acid metabolism and energy production in the body. Its role in supporting the adrenal cortex is based on its involvement in the biosynthesis of steroid hormones, including cortisol, which are produced by the adrenal glands. Animal studies from the mid-20th century demonstrated that severe pantothenic acid deficiency could impair adrenal function and reduce the ability to produce corticosteroids. However, such deficiencies are extremely rare in humans, and overt adrenal insufficiency from B5 deficiency has not been documented in clinical settings.

Some small studies and anecdotal evidence suggest pantothenic acid supplementation may help support adrenal health, particularly under stress, but controlled human trials are lacking. The mainstream scientific consensus is that, while pantothenic acid is required for normal adrenal function as part of overall metabolism, there is limited direct evidence that supplementation above the recommended dietary allowance provides specific benefits to the adrenal cortex in healthy individuals.
