Hericium mushroom, also known as Lion's Mane (Hericium erinaceus), has gained attention for its potential effects on cognitive function and the cholinergic (acetylcholine) system. Several preclinical studies, particularly in animal models, have shown that extracts of Hericium erinaceus may enhance nerve growth factor (NGF) expression and promote neurogenesis, which could indirectly support acetylcholine signaling by fostering the health of cholinergic neurons. Some research indicates that compounds in Lion's Mane, such as hericenones and erinacines, can increase NGF levels in the brain, which is critical for the maintenance and survival of neurons that produce acetylcholine.

A few small human clinical trials and case reports have shown cognitive benefits in older adults with mild cognitive impairment when supplementing with Lion's Mane, but direct evidence linking Hericium to enhanced acetylcholine levels or function in humans is limited. Most of the support comes from preclinical findings, with human data still emerging and somewhat preliminary.

In summary, while there is some scientific basis for the use of Hericium mushroom to support the acetylcholine system through neurotrophic effects and potential cognitive benefits, the quality and quantity of direct evidence in humans are not robust. Therefore, the scientific validity exists but is currently limited, warranting a moderate-low evidence rating.
